纳米医药技术以介观量级尺寸对材料的分子(组装)结构进行分析和操控,是纳米技术与生物学的结合,在生物医学领域应用,具有对单个或多个生物活性分子进行时空精准探测、修饰、操控、运输等潜力,从而为分子生物研究提供强大的解析工具、对重大疾病实现高精度监测和精准的时空调控, 尤其微纳米医用材料在药物定点定位递送&控释、基因编辑、生物微环境监测等方面具有重大的应用前景。

随着中国逐步进入老龄化社会,根据《国家创新驱动发展战略纲要》,发展安全、有效、便捷的健康技术,应对人口老龄化的挑战,是生物医学研究的重点方向。微纳米生物材料是国际生物技术领域的前沿和热点,在医药卫生领域有着广泛的应用和明确的产业化前景,特别是纳米药物载体、纳米生物传感器和成像技术以及微型智能化医疗器械等,在疾病的诊断、治疗和卫生保健方面发挥重要作用。美国、日本、德国等国家均已将纳米生物技术作为21世纪的科研优先项目予以重点发展。目前我国生物医疗材料产业以传统材料工艺为主,在高性能的微纳米生物材料发展上落后于上述发达国家,医疗产业的微纳米生物材料引进为主,虽然近年来在材料基础研发上取得了阶段性进步,然而微纳米生物材料的产业化水平仍较为薄弱,是我国生物医药产业达到国际先进水平的主要瓶颈。

纳米医药研究室研究室,隶属于大连理工大学生物医学工程学院,以纳米医用材料研究为核心,目前的研究方向包括1)基因药物递送系统的开发和应用:通过设计合成安全高效的基因递送功能分子,构建了包载DNA的纳米输运载体,基于对DNA凝缩理论的系统和深入研究,实现了对包载凝缩DNA纳米运载体的尺寸、形状和形状的精准控制,在病灶部位实现了持续高效的表达药用蛋白。2)克服血脑屏障的高渗透药物递送系统:针对脑退化疾病(阿尔海默病、帕金森病等)开发高效通透血脑屏障的药物递送系统,实现靶向输送药物到神经元细胞或胶质细胞。3)肿瘤高靶向性、高渗透性纳米药物递送系统的研究:构建多功能、环境响应型、靶向性、智能化的聚合物纳米药物递送系统,通过对聚合物高分子的分子结构及其组装的纳米精准控制,实现对各类恶性肿瘤实现高效的治疗效果。4)具有响应性,功能性的生物水凝胶:针对人类重大慢性病(如眼科疾病:白内障,葡萄糖膜炎等疾病)以及人类重大疾病(原发或复发性恶性癌症等)开发的的高度可控的模块化介入型的纳米药物缓释体技术。目前微纳米生物材料研究团队在肿瘤等重大疾病的早期精准诊断和靶向治疗领域取得了突出的进展,近5年在国际著名学术期刊Nature Nanotechnology、Nature Communications、ACS Central Science、ACS Nano、Chemistry of Materials、Biomaterials、Small、Journal of Controlled Release、Molecular Therapy等发表论文60余篇、10余项发明专利以及针对高阶脑胶质瘤和眼部慢性病2项临床转化技术。

未来纳米医药研究室将注重研究深度和产业转化的提升,基础研究的重点包括:精密表微观结构的纳米功能/智能材料的可控制备、纳米生物材料的分子设计及其结构控制及其与生物分子及细胞之间的相互作用、纳米控释给药系统、纳米分子影像探针构建以及纳米材料器件与分子组学等纳米生物医学方向;产业化重点以威胁人类健康的重大疾病(包括:癌症、中枢神经退行性疾病)为目标,推动发展靶向给药纳米技术、肿瘤微环境纳米药物、精准医学纳米技术、肿瘤免疫治疗纳米药物等平台技术,为国家全面迈入小康社会提供有效的医疗保障。
